Monoclonal Antibodies to Identify Tomato Mosaic Tobamovirus (tomv)
Monoclonal antibodies were obtained against Tomato mosaic tobamovirus (ToMV) isolated in Brazil. One antibody (8G7G2) isotyped as IgG2b (κ light chain) showed strong specificity and very low cross reaction with the Tobacco mosaic virus (TMV). It can be used in identification of tomato mosaic virus (ToMV).
متن کاملPhosphorylation and/or presence of serine 37 in the movement protein of tomato mosaic tobamovirus is essential for intracellular localization and stability in vivo.
The P30 movement protein (MP) of tomato mosaic tobamovirus (ToMV) is synthesized in the early stages of infection and is phosphorylated in vivo. Here, we determined that serine 37 and serine 238 in the ToMV MP are sites of phosphorylation. MP mutants in which serine was replaced by alanine at positions 37 and 238 (LQ37A238A) or at position 37 only (LQ37A) were not phosphorylated, and mutant vir...
متن کاملExpression of Genetically Controlled Resistance to Tobacco Mosaic Virus Infection in Isolated Tomato Leaf Mesophyll Protoplasts
Phenotypic expression of different genes controlling resistance to tobacco mosaic virus (TMV) in tomato was analysed in the protoplast system using otherwise isogenic breeding lines. Genes Tm-2 and Tin-22 were not expressed and did not prevent TMV-L, a common tomato strain of TMV, infecting and multiplying. By contrast, homozygous gene Tin-I was able to express its effect in protoplasts as well...
متن کاملExpression of the tomato ringsport nepovirus movement and coat proteins in protoplasts.
Tomato ringspot nepovirus (TomRSV) produces a 45 kDa movement protein and a 58 kDa coat protein in infected plants. Accumulation of the movement protein in relation to that of the coat protein was studied in infected protoplasts using a monoclonal antibody against the movement protein and polyclonal antibodies against the coat protein. Unlike most other viral movement proteins, the TomRSV movem...
متن کاملTwo amino acid substitutions in the tomato mosaic virus 30-kilodalton movement protein confer the ability to overcome the Tm-2(2) resistance gene in the tomato.
The Tm-2(2) resistance gene is used in most commercial tomato cultivars for protection against infection with tobacco mosaic virus and its close relative tomato mosaic virus (ToMV). To study the mechanism of this resistance gene, cDNA clones encompassing the complete genome of a ToMV strain (ToMV-2(2)) that was able to break the Tm-2(2) resistance were generated. Chimeric full-length viral cDNA...
